In vitro culture protocols of a selected cultivar of Lycium barbarum L., aimed at the conservation of germplasm and micropropagation for nursery purposes

Abstract
Within the scope of the project to enhance and safeguard the production chain of the goji in Italy under the GOJI ITALIANO® trademark, a Working Group was created to develop an effective protocol for the in vitro propagation of goji.
The initiative stems from the need to preserve the germplasm of ecotypes of goji, from which some cultivars have been selected by the specialized farmers of the Network of Enterprises “Lykion”, in primis within the field-catalog of Taurianova (RC), planted in 2014 and later, since 2015, distributed in orchards of the provinces of Calabria (except Catanzaro) and of other regions of southern Italy and, finally, the need to maximize the nursery reproduction of young plants for new fruit-growing plant.
The attention has been turned to selected cultivars taking 30-40 cm of scions during the vegetative rest from plants originated from a field in the municipality of Sant’Ilario dello Ionio (RC). At the Micropropagation Laboratory of the Seed Bank of HortiVeS, specialized in the safeguard and study of local horticultural varieties, food plants of the regional tradition and also wild edible flowers and herbs (NIGRO et al., 2017), several sterilization protocols have been evaluated with particular attention to the pre-treatment phases of stock plant materials (scions), aimed at minimising the level of environmental pollution of the explants.
The substrates used for the adaptation phase to in vitro culture were based on Murashige and Skoog (1962) and Murashige and Skoog (1962) modified.
After a month from the cultivation of the scions, between 80 and 100% of the buds sterilized with sodium hypochlorite were found decontaminated and vital.. However, we could observe that in some cases, even the explants that were initially vital, later suffered phenomena of apical necrosis, tissue dehydration, vitrescence and callus formation.
In order to overcome the criticalities found, we will proceed by setting up new experiments, paying attention to the application of sterilization protocols aimed at minimizing the oxidative stress of the sterilizing agents (NaClO 1%) and the substances utilized for the pre-treatments (C2H6O 80%).
