Abstract
Grapes represent a vital beverage crop in South Africa, continually threatened by diseases like Botrytis cinerea. Indigenous grapevine-associated microbes offer unexplored opportunities for sustainable biocontrol.
This investigation profiled microbial communities from various Vitis vinifera cultivars and stone fruit in Stellenbosch, South Africa and from a vineyard in the Lombardy region of Italy.
Endophytic and epiphytic yeasts and bacteria were isolated using various selective and differential media.
Antifungal activity against Botrytis cinerea was tested via in vitro and in vivo assays.
Various yeasts and bacteria including Aureobasidium pullulans, Filobasidium oeirense, Hanseniaspora spp., Metschnikowia spp., Pichia kluyveri, Suhomyces prunicola, Klebsiella spp., Pantoea spp., and Pseudomonas spp. were identified.
The production of cell wall degrading enzymes (glucanases and chitinases), as well as biofilm formation were found to be common features among the isolates with antifungal activity, while the production of volatile organic compounds appeared to have limited contribution to ability of the isolates to inhibit B. cinerea mycelial growth.
Intraspecific variation was evident within species such as Pantoea agglomerans. The study revealed a wide diversity of Metschnikowia spp. including M. hawaiiana, M. reukaufii and M. chrysoperlae whose antifungal activity has not been extensively studied, thereby opening opportunities for further evaluation.
