Abstract
A biolistic transformation procedure for lily hasw been developed in order to establish resistance against lily symptomless virus (LSV). A plasmid bearing the hygromycin phosphotransferase gene, the
-glucuronidase (GUS) marker gene and the coat protein gene from LSV fused to the 35S-promoter was introduced into bulb scale tissue by microprojectile bombardment.
The bombardments were performed at different intervals after induction of cell division in the bulb scale tissue in order to find optimum delivery efficiency.
Selection of the bombarded tissue for four months produced hygromycin resistant calli.
Most of these calli (95%) were GUS+. From the GUS+ calli, shoots and bulblets were regenerated and analyzed for the expression of the GUS marker gene and the coat protein gene of LSV.
