Abstract
Genetic relatedness among 16 phytoplasma isolates from various sources was studied by different molecular techniques including polymerase chain reaction (PCR)-based procedures using universal and specific primer pairs and restriction fragment length polymorphism analysis.
The work has resulted in the identification of new isolates as aster yellows (AY) phytoplasmas.
DNA heteroduplex mobility assay (HMA) and perpendicular temperature gradient gel electrophoresis analyses were performed as additional molecular techniques in order to assess their sensitivity, reliability and simplicity for detection and differentiation of phytoplasma isolates.
HMA combined with PCR has been shown to be a very useful method for detection and differentiation of phytoplasmas.
