Abstract
The origin of variation associated with anther culture was examined using two varieties, RPKN2 and BW 306–8. Variation was partitioned into among doubled haploid (DH) lines, among regenerants, among tillers in a regenerant and among plants in a tiller (plants derived from a DH line). Significant variation among DH lines was observed in seven characters for RPKN2 and in one character for BW 306–8. Among regenerants, variability was observed in two traits for both RPKN2 and BW 306–8. Variation was likewise detected among tillers within regenerant for some traits in both materials.
Plants in a tiller within a regenerant (pooled variation) also differed significantly in a number of characters.
The variation observed among regenerants of an anther indicates that different mutations occurred during callus formation, proliferation and/or organogenesis.
Variation among tillers in a regenerant suggests that a regenerant may have originated from different cells with different mutations.
Variation among plants in a tiller could be due to genetic segregation, implying selection within lines should be employed aside from selection among lines.
This study provides an insight into the magnitude of variability that can be observed within doubled haploid derived purelines.
