Abstract
Growth and development of Limonium spp. cv.
Misty Blue and Fragaria x ananassa cv.
Houkouwase plantlets cultured in Tosilee mix, vermiculite, or perlite contained in plug cells were evaluated against those in an agar medium.
The culture solution initially added per plant for statice and strawberry were 15 ml MS and 10 ml half strength MS medium, respectively, with no added sugar or vitamin.
Explants were cultured in a room with cool white fluorescent lamps, 16 h photoperiod, 25±1°C air temperature, 70–80% RH, and 1000 ppm CO2 during the photoperiod.
A 10 mm round hole on the vessel lid sealed with a microporous filter raised the number of air exchanges per hour of the vessel to 2.8. Fresh weight, height, root length, and chlorophyll concentration of the statice plantlets, and height, root length, and shoot-root ratio of strawberry were significantly increased in Tosilee mix.
The agar medium gave the poorest results.
