PLANT REGENERATION FROM LEAVES OF ULMUS X ‘COMMELIN’

For genetic transformation, an efficient plant regeneration system is required.
We report here on plant regeneration from leaf discs of the Dutch elm hybrid ‘Commelin’. Buds were induced after 2–3 weeks near ribs at the edge of leaf discs excised from micropropagated plants.
Bud initiation was promoted by the use of agarose instead of agar and diluted Murashige-Skoog (MS) medium (half strength). It was maximum when thidiazuron (TDZ; 0.5 or 0.1 μM) was combined with indoleacetic acid (IAA; 0.06 or 0.6 μM). The best results (10–12 buds/explant) were obtained with maltose or fructose (55 mM) used as sugar.
Shoots developed within 5–7 weeks when TDZ was lowered (0.01 μM) or replaced by benzylaminopurine (BAP; 1 μM). Elongation occurred for buds initiated on maltose or sorbitol and transferred on a maltose medium (27.5 mM). The use of unsealed Petri dishes or high Petri dishes slightly improved adventitious bud formation when IAA was supra-optimal.
Shoots were rooted on Driver-Kuniyuki (DKW; ½ strength) medium containing indolebutyric acid (IBA; 2.5 μM) and successfully acclimatized.
