Abstract
To date there is relatively little DNA sequence information from Allium. We have sequenced cDNAs from A. cepa libraries prepared from bulbs, roots and seedlings to identify loci on the A. cepa linkage map and facilitate development of PCR-based expressed sequence tag ( EST) markers.
BLAST comparisons with GENBANK sequences indicated 80% of these showed homology with genes of known function or ESTs.
Eighteen of sequenced clones were mapped as restriction fragment length polymorphisms (RFLP) and were of abundant transcripts encoded by gene families (CAB proteins, RuBISCO, ubiquitin, elongation factors). EST markers are being developed from selected sequences.
