PHYLOGENETIC CLASSIFICATION OF INTRODUCED AND INDIGENOUS ACTINIDIA IN JAPAN AND IDENTIFICATION OF INTERSPECIFIC HYBRIDS USING RAPD ANALYSIS

Abstract
RAPD analysis was used to examine the phylogenetic relationships amongst four introduced and indigenous Actinidia species in Japan, A. deliciosa, A. chinensis, A. rufa and A. arguta and some interspecific hybrids.
In all, 21 cultivars or selections were studied.
A total of 181 fragments were generated by 14 selected RAPD primers, and 165 bands were found to be polymorphic in the cultivars or selections used.
A dendrogram based on these band patterns clearly separated into four clusters, consisting of A. deliciosa, A. chinensis, A. rufa and A. arguta. It was shown that A. deliciosa and A. chinensis were closely related, whereas A. arguta was relatively distant from the other species. A. arguta and A. rufa have traditionally been considered as close relatives, but this was not supported by RAPD analysis.
The interspecific hybrids, ‘Shinzan’ (reported to be A. arguta × A. deliciosa) and ‘Sanryoku’ (reportedly A. deliciosa × A. chinensis) had high similarity coefficients with their respective parental species and in the dendrogram, ‘Shinzan’ and ‘Sanryoku’ clustered with their respective maternal species. ‘Kosui’ (reportedly A. arguta × A. deliciosa) had considerably lower similarity coefficients with its putative parents.
In the dendrogram, ‘Kosui’ did not cluster with either A. arguta or A. deliciosa but did with A. rufa, suggesting that A. rufa may be one of its parents.
