ALLOZYME VARIATION OF A CRYPTOMERIA JAPONICA PLANTATION IN LIU-KUEI BRANCH, TAIWAN FORESTRY RESEARCH INSTITUTE

Abstract
Using starch gel electrophoresis, we analysed allozyme variation of one-year-old leaf tissue of 1,000 individuals from a Cryptomeria japonica plantation.
A total of 100 seed-lots was introduced from Japan and the plantation established in 1973 at Liu-Kuei Branch, Taiwan Forestry Research Institute. Skdh-1 and Mdh-1 were monomorphic, Skdh-2, Mdh-2, Aat-1,and Aat-2 have 2 alleles each.
The average allele number per locus of 3 enzyme systems (Skdh, Mdh, and Aat) was 1.37, and the expected heterozygosity (He) was 0.152 which is lower than reported values: 0.189 or 0.178, based on natural populations in Japan.
Thus it is concluded that the 100 seed-lots may have originated from a limited number of individuals from a natural population in Japan.
Differentiation between seed-lots (Fst) was 0.07, therefore most of the variation was attributable within the seed accession.
