ROOTING AND ACCLIMATIZATION OF IN VITRO PROPAGATED OLIVE TREE MICROSHOOTS (OLEA EUROPAEA SATIVA L.) CV. CHONDROLIA CHALKIDIKIS

Microshoots of the Greek olive cv.
Chondrolia Chalkidikis were cultured in vitro in WPM supplemented with 4 µΜ zeatin.
The effect of various auxin concentrations (IBA, NAA) on root induction was studied.
Rooting percentage up to 70% was achieved by the combination of 12 µΜ ΙΒΑ + 3 µΜ ΝΑΑ although abscission of shoot tips and leaves was common.
The effect of 30 &degmicro;Μ putrescine and 100 ppm H₃PO₄ addition to the medium was also tested.
The percentage of rooted microshoots as well as the number of roots/microshoot were both increased by the putrescine treatment reaching up to 93% and 4%, respectively.
Root length was reduced by the addition of phosphoric acid.
Rooted and non-rooted shootlets were planted in peat:perilte 4:1 (by volume) under 90% RH. Survival of rooted microshoots was high (90%) while that of non-rooted ones was low (30%). Furthermore, ex vitro trials resulted in no rooted plantlets
