Abstract
Induction, maturation and germination of somatic embryos of jojoba [Simmondsia chinensis (Link) Schneider] from leaf tissue of female plants cultured in vitro were obtained.
Leaf explants were placed on their adaxial sides in Petri dishes and maintained in darkness on the half-strength Murashige and Skoog basal medium (MS/2). The use of 2,4-dichlorophenoxyacetic acid (2,4-D, 4.52 µM) with 3 cytokinins: 6-benzylaminopurine (BAP, 4.43 µM), N-(2-chloro-4pyridyl)-N’-phenylurea (CPPU, 4.03 µM) or (E)-6-[3-(trifluoromethyl)-but-2-enylamino]purine (F3iP, 3.71 µM) led to the formation of embryogenic calli and somatic embryos.
After two 30-day subcultures, embryogenic calli were transferred onto MS/2 medium supplemented with different auxins and cytokinins.
Somatic embryo maturation and conversion into in vitro plantlets were obtained.
The histological study has shown the conformity of each stage.
A correlative study was carried out to focus the effect of growth regulators, age of explants and free polyamine content during embryo induction stage.
