ADVENTITIOUS SHOOT REGENERATION FOR AGROBACTERIUM-MEDIATED TRANSFORMATION IN PRUNUS

Abstract
An efficient regeneration protocol has been defined to induce organogenesis from micropropagated shoots of the plum rootstock Ferdor (Prunus insititia x domestica), some peach [P. persica (L.) Batsch] genotypes and a cherry cultivar (P. avium L.). Apices were excised from in vitro shoot cultures of peach and plum and cultured 21 days in dark on a LP medium supplemented with BA and NAA, and then transferred to an auxin free medium in the light for 21 days.
The first apical leaves were excised from these apices and cultured in the same way.
During dark incubation leaves developed a callus at the petiolar base.
Adventitious shoots appeared on this callus after transfer to auxin free medium in the light.
A shorter protocol was applied to the shoot apices of Lapins cherry in order to evaluate the regeneration response.
Adventitious shoot regeneration was also achieved from hypocotyl slices of plum.
These regeneration systems are going to be used for experiments of Agrobacterium-mediated genetic transformation.
