THE INFLUENCE OF SHORT-TERM COLD STORAGE ON THE CHERRY MICROSHOOT PROLIFERATION

Abstract
The aim of this research was to evaluate the influence of short-term cold storage (6 weeks at +4°C) on the microshoot proliferation rate, length and rooting.
The number of surviving microplants after planting ex vitro was enumerated.
Five promising cherry cultivars were chosen for the evaluation.
Rooted cherry microplants were used for the experiments.
The vitality and proliferation rate of microshoots after cold storage (C) was significantly higher than for non-stored (NC) microcultures.
During four subcultures, length of microshoots fluctuated for both microculture types, however C in vitro cultures had longer microshoots, increased percentage of rooted microshoots, and better adaptation into non-sterile conditions.
We conclude that short-term cold storage has positive influence on cherry microshoot proliferation, rooting, and establishment ex vitro in spite of a six weeks non-propagation period.
