Abstract
Squash technic regularly used in Prunus species do not permit a precise study of mitotic chromosomes morphology.

A new method with dividing protoplasts may allow development of banding processes for an accurate identification.

Protoplasts, like lymphocytes used in animal cytogenetics, are submitted to an hypotonic shock before spreading on a slide.

After drying and Giemsa colouring, heterochromatin appears very clearly in Prunus species studied.
