GENETIC VARIATION OF JIAOGULAN (GYNOSTEMMA PENTAPHYLLLUM) CULTIVAR ‘SIBSONGPUNNA 1’ REVEALED BY RAPD AND ISSR MARKERS

Abstract
Jiaogulan (Gynostemma pentaphyllum) cultivar ‘Sibsongpunna 1’ was introduced from Southern China.
It was propagated by seeds which might cause variations within the cultivar.
The identification of promising clone based on morphological characters can be difficult and complicated.
Our objective was to examine the genetic variation by using Randomly Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeat (ISSR) markers.
Based on the DNA polymorphic bands generated, 30 primers were selected to identify each accession.
The 30 primers produced 607 fragments ranging in size from 126 to 2739 base pairs.
Of these, 542 fragments were polymorphic and only 65 are monomorphic indicating high genetic variation within the cultivar.
All polymorphic fragments were used to calculate a similarity matrix and to perform UPGMA cluster analysis.
The result revealed that genetic relatedness among 20 accessions was in the range of 50-100% and could be divided into three groups.
Each accession had unique DNA profile suggesting that the technique used in this study was able to detect plant to plant variation and was effective for clonal selection.
