TESTING OF RESISTANCE TO ERWINIA AMYLOVORA IN AN IN VITRO CULTURE ASSAY

Abstract
Fire blight, caused by Erwinia amylovora, is an infectious disease affecting pome fruit and other members of the rose family.
It is a serious concern to producers of apples and pears in the Czech Republic.
Under optimal conditions, it can seriously damage an entire apple or pear orchard in a growing season.
Resistance of old local pome fruit cultivars to E. amylovora was tested in an in vitro culture assay.
Four pear cultivars (‘Belle de Bois’, ‘Jakubka ceska’, ‘Doyenne de Merode’, ‘Solanka’) and two apple cultivars (‘Boiken’, ‘Nonnetit’) were selected for testing.
A verified isolate of E. amylovora (Ea 8/95) isolated from necrotic tissues of hawthorn (Crataegus × monogyna) was used in tests.
The apical tip of the shoot was removed with scissors and two concentrations of the pathogen (103 and 106 cfu/ml) were applied on the cut surface.
Based on the results, ‘Solanka’ was the most resistant and ‘Doyenne de Merode’ the most susceptible of the selected pear cultivars.
Concerning apple, two cultivars ‘Boiken’ and ‘Nonnetit’ were tested.
In this test, ‘Boiken’ was three times more susceptible than ‘Nonnetit’. ‘Nonnetit’ showed minor symptoms of necrosis 9 days after inoculation.
Preliminary results from inoculation of pear and apple shoot tips maintained in tissue culture with E. amylovora was a useful method to test fire blight resistance in pome fruit cultivars.
The obtained results are preliminary and evaluation of additional cultivars will be carried out in following years.
