CHLOROPLAST SIMPLE SEQUENCE REPEAT (CP-SSR) ANALYSIS FOR DETERMINING THE ORIGIN OF J. HOPEIENSIS

Abstract
To define the source of Ma walnut (J. hopeiensis), 10 chloroplast simple sequence repeat (cp-SSR) primers were used to amplify SSR fragments from
7 samples of J. regia, 8 samples of J. mandshurica, 34 samples of J. hopeiensis, and 1 hybid of J. mandshurica♀×J. regia♂. The results showed that four cpSSR loci were polymorphic.
According to alleles of all samples generated by the four polymorphic cpSSR loci, the genotypes of the hybrid of J. mandshurica♀×J. regia♂ and 24 samples of J. hopeiensis were the same to that of J. mandshurica. Only 1 sample of J. hopeiensis named ‘Yanqingmahetao-2’ had the same genotype as J. regia. The genotypes of 9 samples of J. hopeiensis from Shaanxi were the same as that of J. mandshurica in 3 polymorphic cp-SSR loci, while they were the same as that of J. regia in 1 polymorphic cp-SSR locus.
These results indicated that J. hopeiensis originated mainly from the natural hybrid of J. mandshurica♀×J. regia♂. while 97.1% of J. hopeiensis came from the hybrid of J. mandshurica♀×J. regia♂, while 3% of J. hopeiensis came from J. regia♀×J. mandshurica♂ in the wild population of J. hopeiensis. Furthermore, genetic difference in cp-SSR existed between J. hopeiensis from Shaanxi and those from Beijing and Hebei.
