Induction and proliferation of durian (Durio zibethinus) embryonic culture in media supplemented with auxin and cytokinin

Abstract
Durian is one of Indonesia’s main fruit commodities that have high economic value.
In vitro somatic embryogenesis can be used as an alternative way to provide clonal rootstocks, as well as for biotechnology-based plant improvement.
This research consisted of three experiments and used mature zygotic embryos as explants.
The first experiment was callus induction on a combination of Murashige and Skoog (MS) medium, woody plant medium (WPM), or Gamborg’s B5, and picloram (0, 1, 2, 3, 4 and 5 ppm). The second experiment was callus proliferation and induction of embryogenic callus that used several different media: media from the previous stage, and MS + BAP 1 ppm + picloram (3, 4 and 5 ppm). The third experiment was maturation of somatic embryos on MS medium to which an auxin (NAA (0, 1, 2, 3 and 4 ppm) or picloram (0, 4, 8 and 12 ppm)) and BAP (0, 1 and 2 ppm) were added.
Calli were successfully induced in all treatments, and the percentage of callus reach 100% in some treatments from the first experiment.
The highest callus proliferation based on callus weight occurred in MS medium with 1 ppm BAP and 3 ppm picloram at the fourth subculture.
Embryogenic callus could be induced in all media combinations.
Globular pro-embryos could be induced in all treatments of MS medium with the addition of BAP and NAA, but none underwent maturation.
