Phylogeny of Indonesian durian (Durio sp.) germplasm based on polymorphism of ITS-nrDNA sequences

Abstract
Indonesia is considered as the center of origin of durian, and has abundant species and cultivars.
Phylogenetic analysis is one of the methods to study the genetic structure of germplasm for management purposes.
This research aimed to analyze the molecular phylogeny of Indonesia’s durian germplasm based on polymorphism of conserved ITS-nrDNA region.
One hundred and forty-nine durian germplasm samples were used in this study.
ITS sequences of the germplasm samples and those from GenBank were aligned using Clustal W, and a phylogenetic tree was reconstructed using Geneious R6 PC software.
The results demonstrated that the ITS-nrDNA region is highly conserved in genus Durio and is a powerful tool to determine durian systematics at the interspecies level.
The phylogenetic tree could be drawn with speciation of several doubtful accessions and new emerging durian variations could be confirmed.
The tree conveyed low evolutionary resolution of the region around Durio zibethinus; therefore, this gene is not suitable for identification of genetic diversity of durian at the intraspecies level in this species.
