Phenotypic behavior and assessing phage susceptibility in pathogenic Xanthomonas arboricola pv. juglandis

Abstract
Xanthomonas arboricola pv. juglandis (Xaj) associated with walnut blight, the most significant aboveground bacterial disease causing tremendous loss worldwide.
Prospective orchards between 2021 and 2022 in Hungary and Romania showed that the disease continues to spread geographically and affects several cultivars: Juglans regia ‘Sebeshelyi gömböly’, ‘Milotai 10’ and ‘Alsószentiváni 117’. Isolation resulted in 24 putative Xanthomonas spp. and their chemotaxonomic analysis using MALDI-TOFMS assigned seven of them in the arboricola species.
Interestingly all these seven strains were resistant to both copper and streptomycin at 10 µg mL‑1. HU4, belonging to morphotype B, was able to tolerate 25 µg mL‑1. A set of tests using commercial API kits revealed a high variability in biochemical proprieties.
Phage susceptibility profiles support this polymorphism and strain dependent criteria.
This study investigated the population variance and resistance in Xaj to provide an improved understanding of pathogen populations associated with walnut blight and to lead to an effective control measure.
