Abstract
Fluorescent staining technics are described for detection of: a) pollen grain and pollen tube nuclei using the fluorescent DNA probes DAPI, and: b) pollen tube walls with both aniline blue and calcofluor (fluorochromes which stain more or less specifically the callose). Detection of vegetative and generative nuclei was possible with flow cytometry of isolated pollen.
Growth cinetic of pollen tubes was studied during the 24 hours following artificial pollination.
These techniques, applied to several Rosa cultivars and species are described, illustrated and discussed.
