KONJAC MOSAIC VIRUS, DASHEEN MOSAIC VIRUS AND UNKOWN POTYVIRUSES INFECTING ZANTEDESCHIA SPP. AND OTHER CULTIVATED ARACEAE

Abstract
Electron microscopical examination of symptomatic Zantedeschia aethiopica and of Zantedeschia sp. hybrids, Amorphophallus spp., other tropical Araceae and Arum italicum revealed infections with potyvirus-like particles.
In several of these plants Dasheen mosaic virus (DsMV). was identified by immunoelectron microscopy (IEM). In addition, many plant samples revealed potyvirus-like particles which did not react with DsMV antiserum.
While virus particles in some samples reacted strongly with antiserum against Konjac mosaic virus (KoMV), further potyvirus-like particles were found in Zantedeschia not reacting with either of the antisera against DsMV or KoMV. By IEM decoration at least four distinct potyviruses were found infecting Araceae.
Mixed infections with these viruses occurred frequently.

Comparative decoration tests revealed that DsMV and KoMV are serologically clearly distinct viruses with only a relatively distant serological relationship.
As demonstrated by decoration titer assays using antisera against both viruses, heterologous reactions reached 4 to 9 steps lower decoration end points than decorations of the respective homologous viruses.
Decoration tests using a panel of antisera against a large number of different potyviruses revealed only weak serological relations with DsMV and KoMV.

Sequence analysis of the 3‘ terminal regions of the KoMV genome, revealed the closest relationship of KoMV with Japanese yam mosaic virus and a more distant relationship with DsMV, confirming the classification of each virus as a distinct species of the genus Potyvirus. The identity of the remaining potyviruses found in Zantedeschia however still remained elusive.

For routine potyvirus detection in Zantedeschia a broad spectrum genus specific monoclonal antibody was utilized in an ACP- ELISA format.
It could be demonstrated that all potyviruses found so far, including DsMV and KoMV, are detected in infected plants, however there were stronger ELISA reactions with DsMV infected materials.
