Abstract
The evergrowing phenotype of peach [Prunus persica (L.) Batsch] is controlled by a recessive allele in a single gene (evergrowing or evg) and is a possible model system to study winter dormancy and cold-hardiness.
We report here the progress towards developing a physical map at the evg gene region using a newly constructed peach BAC library.
Three separate contigs were developed at three AFLP marker positions that were close to evg on the genetic linkage map.
To confirm the correspondence of the contig positions to the AFLP markers, simple sequence repeat (SSR) markers were developed from peach BAC clones and genetically mapped in this region.
In order to discover the BAC clones containing the evg gene, a chromosome walk in both directions was initiated from the BAC clone PpN18F12, which contains the AFLP marker that is closest to the evg locus (1 cM mapping distance). This contig has been expanded to 20 clones.
Genetic mapping of the SSR marker, pchgms29, developed from the extended BAC clone, PpN109L12, indicated that the direction of the chromosome walk towards the evg locus was correct.
