IN VITRO ROOTING STUDY OF THE PEACH-ALMOND HYBRID MAYOR®

Abstract
The optimal in vitro conditions to induce explant rooting of the ‘Mayor’® peach-almond hybrid were determined by comparing the different auxinic concentrations, methods of treatment and culture media.
In the first experiment three IBA concentrations were studied: 1, 1.5 and 2 mg/l using the in vitro culture medium created specifically for this hybrid, according to the leaf macronutrient concentration, by our research team, at half of it’s normal concentration.
As a gelifying agent we used 7 g/l of BactoAgar Difco, 15 g/l of sucrose and adjusted the pH to 5.7. After 25 days of culture the best IBA concentration was 1 mg/l with a 20% success rate of rooting explants.
In order to improve this low rooting percentage, vermiculite was included in the culture media, with the purpose of changing the physical properties.
Four vermiculite treatments were studied: media concentrations (60:40, 60:60, 60:80 and 60:100) (v:v), using 2 mg/l of IBA as a growth regulator.
The results in this experiment were better than the previous ones, with a 79,1% rate of rooting in 60 ml of vermiculite:100 ml of culture media.
With the culture media optimised, five IBA concentrations (1, 1.5, 2, 2.5 and 3 mg/l) were added to the culture media for comparison, and three dipping processes of the explant bases in an IBA concentrate solution (500, 750 and 1000 mg/l) were studied.
The results obtained showed that, following dipping in 750 or 1000 mg/l, a 100% success rate of rooting explants was obtained, whereas the best inclusion in the culture media was 2 mg/l with a 73,6% success rate.
These results indicate the greater effectiveness of the dipping process, in the percentage of rooting as well as in the number and length of the roots produced.
