RUNNER CERTIFICATION AND VIRUS ELIMINATION IN COMMERCIAL STRAWBERRY CULTIVARS IN AUSTRALIA

Abstract
An extensive strawberry virus indexing program run by Crop Health Services (CHS) (Department of Primary Industries Knoxfield), provides runner certification to the Australian strawberry runner industry on an annual basis.
Biological indexing by petiole grafting onto sensitive indicator species is the basis of the certification pro-gram.
The grafting assay reliably detects Strawberry mottle virus (SmoV), Straw-berry crinkle virus (SCV), Strawberry mild yellow edge virus (SMYEV), Strawberry vein banding virus (SVBV), Tobacco streak virus (TSV) and Pallidosis.
Confirmatory testing for Strawberry mild yellow edge potexvirus is also done using enzyme-linked immunosorbent assay (ELISA). The runners are indexed for the fungal diseases caused by Phytophthora sp., Verticillium sp. and Gnomonia comari (leaf blotch), and monitored for the presence of phytoplasmas and bacteria.
Elimination of viruses from strawberries is achieved through thermotherapy in combination with meristem culture.
Tissue culture also enables rapid multiplica-tion, which reduces the time to planting when large numbers of runners are required in the field.
The CHS tissue culture laboratory is an Australian Quarantine Inspec-tion Service (AQIS) accredited facility for the tissue culture multiplication of im-ported strawberry varieties while they are under post-entry quarantine conditions.
