STRAWBERRY GERMPLASM CONSERVATION: THE COLLECTION AT IFAPA-CIFA (MÁLAGA, SPAIN)

CIFA (Centro de Investigación y Formación Agraria, Málaga, Spain) has a Fragaria germplasm collection of more than 400 accessions in support of breeding programs.
The germplasm is maintained in seed banks, actively growing plant collections, and in vitro storage.
The advantages of these are discussed.
The need for longer term storage prompted investigations into cryopreservation.
When a standard encapsulation-dehydration process for cryopreservation was used, the best recovery (40-55%) was obtained after drying for 4-6h when water content of the beads was about 23%. Survival and recovery rates varied among genotypes.
When the new encapsulation-dehydration process of Sakai et al. (2000) was applied, where the apices were
simultaneously osmo-protected with a mixture of 2 M glycerol plus 0.4 M sucrose for 1 h, high percentages (up to 60%) of shoot formation resulted.
