PATULIN DETERMINATION FROM APPLE JUICE BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

Abstract
In the Institute of Food Bioresources a method for patulin determination from apple juice by high performance liquid chromatography was developed.
Extraction of patulin was made in acetonitrile from a sample and purification of an obtained extract, using C.U. Patulin columns (MycoSep®228). The obtained solution is evaporated to dryness under nitrogen and re-dissolved.
Patulin is, afterwards, separated on a chromatographic column C18, 150 x 4 mm, 5 μm (high performance liquid chromatograph Thermo Finnigan), eluted in mobile phase and detected on 276 nm, using an UV-VIS DIODE ARRAY detector.
An internal study for the validation of the method for patulin determination from apple juice, by high performance liquid chromatography was achieved.
In a concentration range from 10 μg/L to 400 μg/L the average recuperation is 89.64%. the detection limit (LOD) is 3.71 µg/L, and the quantification limit (LOQ) is 7.42 µg/L.
