Abstract
Traditional chemotherapy drugs such as camptothecin™ work effectively in destroying cancer cells; however these methods are routinely associated with side-effects.
Research conducted by our group and others using carotenoids such as lycopene and lutein have been shown to act at low substrate levels through the action of intrinsic cytokines and growth factors and their receptors within tissues such as prostate and breast.
Recent work conducted in our laboratory using various plant derived bioactive molecules (PBAM) from fruits and vegetables, in combination with much lower concentrations of traditional chemo have demonstrated significant inhibitory effects on the proliferation of prostate cancer cell lines.
Studies were conducted to determine the effects of a combinatorial therapy using the PBAM Bioflavone-alpha with reduced concentrations of camptothecin™ on the human androgen-responsive prostate cancer cell line, LnCap.
In controlled studies, LnCap cells were treated with various concentrations of the traditional chemo alone, the PBAM Bioflavone-alpha alone, and a combination of camptothecin™ at 2 μmol/L with various concentrations of Bioflavone-alpha for 48 hours.
Cell proliferation was assessed using a real-time continuous live cell viability assay system.
Results showed that all concentrations of the combination phytochemotherapy demonstrated significant inhibition of LnCap cell proliferation, while only the 5 and 10 μmol/L concentration of Bioflavone-alpha alone had a statistically significant inhibition of growth.
The combination of camptothecin™ at 2 µmol/L and Bioflavone-alpha at 10 µmol/L had the highest percentage of inhibition of 69% of the control.
Other phytotherapeutic studies were conducted using PBAM capsaicin and curcumin, both known to have bioactivity against certain cancers.
The mechanism of action was studied by measuring c-Jun NH2-terminal kinase (JNK) activity in the LnCap cells.
Results demonstrate that certain PBAM cause an enhancing effect on existing chemotherapeutic drugs at low concentrations and that Bioflavone-alpha further potentiates certain other bioactive compounds by significantly decreasing cell growth, in a selective manner.
