DETERMINATION OF MATING TYPES OF CRYPHONECTRIA PARASITICA ISOLATES COLLECTED FROM AYDIN PROVINCE/TURKEY BY MULTIPLEX PCR

Abstract
This study was conducted to determine the presence and distribution of the mating types, MAT-1 and MAT-2, and vc (vegetative compatibility) groups of Cryphonectria parasitica in the Aydın Province in Turkey.
Total of 100 C. parasitica isolates collected from the most affected nine chestnut growing villages of Nazilli, Sultanhisar, Köşk and Central Counties of the province were used in the study.
Mating types of the isolates were determined by multiplex PCR assay with specific MAT-1 and MAT-2 primers.
DNA fragments obtained from PCR were observed on electrophoresis and evaluated for their molecular lengths.
The fragments which were 1.649 kb and 549 bp in length were identified as MAT-1 and MAT-2, respectively.
Results indicated that 52% of the isolates were MAT-1 and 48% of MAT-2. Only two vc groups (EU-1 and EU-12) were detected.
