DETECTION BY TAS-ELISA OF TOMATO SPOTTED WILT VIRUS NUCLEOCAPSID AND G1 GLYCOPROTEIN IN FRANKLINIELLA OCCIDENTALIS

Triple antibody sandwich enzyme-linked immunosorbent assay (TAS-ELISA) with polyclonal and monoclonal antibodies (PAbs and MAbs) against tomato spotted wilt virus (TSWV) nucleocapsid (N) protein and G1 glycoprotein (GP) was used to detect N-protein and G1 in individual Frankliniella occidentalis. Thrips were collected both within and outside greenhouse pepper crops in Liguria (northwestern Italy) where TSWV infections are severe.
Some thrips were positive for N-protein, but none were positive for G1, irrespective of sex and developmental stage.
More concentrated thrips extracts (25 thrips pooled from a 50% N-protein positive population homogenized in 200 μL) also gave negative results for G1. TAS-ELISA reactions of the two G1 MAbs with TSWV from plants were inhibited in a dose-dependent manner by healthy F. occidentalis extracts but not by aphid extracts; whereas, the reaction of the N-protein MAbs was not affected.
Using the N-protein MAb, it was found that the percentage of TSWV-positive thrips was correlated with infection level in the crop
