NEW TOSPOVIRUSES FOUND IN BRAZIL

Abstract
Surveys conducted in six states of Brazil, by double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), showed that 69 isolates (46%) were tomato spotted wilt virus (TSWV), 54 (36%) were tomato chlorotic spot virus (TCSV), and 12 (8%) were groundnut ring spot virus (GRSV). Fifteen tospovirus isolates (10%) did not show any reaction with the antisera used.
Four candidates for a new tospovirus species isolated from chrysanthemum (Chry 1), zucchini (BR-09z), Tonion (BR-10o), and tomato (BR-11t) were characterized.
The nucleocapsid (N) proteins of all four new isolates were purified and specific antisera were raised.
The ELISA and Western blot analyses showed that they were serologically distinct from the characterized tospovirus species.
Serological comparison among the new isolates showed that Chry 1 and BR-11t were closely related.
Polymerase chain reaction (PCR) amplified fragments from Chry 1, BR-09z, BR-10o, and BR-11t were cloned and partially sequenced.
Southern blot hybridization of the PCR fragments using specific probes showed low homology of the new isolates compared with characterized isolates.
Nucleotide sequence analysis of the Chry1 and BR-09 isolates confirmed results obtained with serology and hybridization assays.
Chry 1 and BR-11t, BR-09z, and BR-10o represent three new serogroups and could be considered new species of the genus Tospovirus.
