Abstract
Somatic embryos of two cultivars of longan (Dimocarpus longan Lour.) Shieryuelongyan and Honghezi, were infected with four wild-type Agrobacterium rhizogenes strains, R1601, R1000, ATCC15834 and MAFF03-01724. A. rhizogenes R1601 showed the strongest virulence.
Twenty days after co-cultivation with R1601, 86.7% of the somatic embryos of ‘Shieryuelongyan’ and 83.3% of those of ‘Honghezi’ had produced numerous hairy roots.
The induced roots varied in morphology.
When the hairy roots were cultured individually on MS basal medium supplemented with 5 mg·l-1 6-benzylaminopurine(BA) and 5 mg L-1 kinetin (KT), somatic embryogenesis occurred directly from the roots of ‘Shieryuelongyan’. The transgenic embryos displayed strong reproducibility of secondary somatic embryos.
However, they developed well on maturation medium supplemented with 0.5 mgL-1 abscisic acid (ABA). Twelve regenerated plantlets were obtained from one independent experiment.
The regenerated plants showed “hairy root” phenotype and a few produced normal leaves.
The transgenic plants were identified by PCR and Southern blot analysis.
