SEGREGATION PATTERNS OF RAPD MARKERS IN AN F1 POPULATION OF LITCHI CHINENSIS Sonn.

Abstract
A F1 population was established from a cross between Litchi chinensis Sonn. cultivars Wuye and Luhebao using the pseudotestcross format.
Twenty-nine 10-mer primers were screened out of 75 primers and 294 RAPD markers were amplified from 68 F1 seedlings.
Of these 53 polymorphic markers, 36 segregated at a ratio of 1:1 (band present; band absent). Among these 36 markers, 21 were present only in ‘Wuye’ and 15 only in ‘Luhebao’; 17 markers segregated at a ratio of 3:1 (band present: band absent) and 232 markers did not segregate in F1 (the parent genotypes may be AA×AA, AA×Aa, Aa×AA, aa×AA or AA×aa). Additionally, 7 markers deviated from Mendelian Law and two showed abnormal segregation patterns.
Of the 146 markers present in two parents, 123 bands (84.2%) did not segregate and 17 bands (11.6%) segregated normally (3:1). Of the 148 markers present in only a single parent, 14.2% were in ‘Wuye’ and 10.2% in ‘Luhebao’. These results provided the necessary data to construct a genetic linkage map of litchi.
