Abstract
Nodal buds excised from severely pruned trees were used for initiation of sterile cultures and their multiplication.
Role of sterilants and antioxidants was also worked out.
Seasonal response and media constitution in different spp. was clearly established for explant establishment and bud induction.
These cultures are being used under in vitro selection scheme employing culture filtrate of F. oxysporum sp. solani.
The second approach employed screening of mesocarp derived callus of P. guajava against the culture filtrate.
Relatively short-term callus cultures were challenged with the toxin contained in culture filtrate.
Preliminary results indicated large scale callus mortality when treated for 30 min.
The use of selection pressure for involving resistance genotype is aimed at increasing the mutation rate in comparison to somaclonal variation.
