MULTIPLE SHOOT INDUCTION FROM SEED DERIVED SHOOTS OF PAPHIOPEDILUM

Abstract
Shoots, derived from seeds, of Paphiopedilum Delrosi were used as explants for multiple shoot induction.
Modified Hyponex medium was supplemented with N6-benzyladenine (BA), thidiazuron (TDZ) or kinetin (Kn) alone and in combination with 2,4-dichlorophenoxyacetic acid (2,4-D) or adenine sulphate.
All explants were cultured for 15 weeks.
It was found that TDZ alone at the concentration of 0.45 μM and 4.52 μM 2,4-D in combination with 0.45 μM TDZ or 4.44 μM BA or 4.65 μM Kn promoted multiple shoots.
High shoot sprouting efficiencies (76.9, 84.6 and 76.9%) and new shoot numbers (1.7, 1.5 and 1.4) were obtained from medium containing 0.45 μM TDZ, 4.65 μM Kn with 4.52 μM 2,4-D and 4.44 μM BA with 4.52 μM 2,4-D, respectively.
Fresh weight, height, numbers of leaf and root of initial plants and new shoots are discussed.
