MODIFICATION OF PHALAENOPSIS METABOLISM BY GENETIC ENGINEERING

Abstract
Phalaenopsis is one of the most important flower crops in Taiwan.
However, the long growth period hampers its commercial production.
In an attempt to increase the metabolic rate and the growth of Phalaenopsis, orchids were transformed with the Vitreoscilla hemoglobin gene via injection of DNA solution into immature capsules.
Putative vhb-transformants were analyzed by Western blotting to verify stable expression of the vhb gene.
The transgenic orchids were characterized for their physiological traits.
Line B47 showed better growth rate (including the vegetative and reproductive stage), had net carbon dioxide assimilation and stored up more malic acid in the dark period.
Line B39 expressed a strong cross-reaction signal by western blotting, and it also had higher root reduction ability.
The results indicated vhb has modified the metabolism of Phalaenopsis.
