MOLECULAR DISCRIMINATION OF “PICOTA” SWEET CHERRIES USING FRUIT TISSUE

Abstract
The aim of this work is to identify molecular markers that allow the differen-tiation of Picota sweet cherries from the Jerte Valley, to establish a reliable method of identification of this type of cherry from fruit samples in the market.
An allele amplified using microsatellite locus UDP98-409 was identified uniquely in the Jerte Valley varieties Ambrunés, Pico Negro and Pico Colorado. This SSR-allele was not detected in other late maturing cherry varieties cultivated in the Valley.
For this work, Prunus microsatellite loci were screened to identify molecular markers specific to these varieties.
Additionally, in order to determine the optimum material to carry out an efficient characterization from cherries in the market, DNA extraction from sweet cherry fruit was optimized using different types of samples such as lyophilized, frozen or fresh fruits.
