ISSR analysis of tree peony (Paeonia sect. Moutan) in Hunan province

Abstract
In order to understand the genetic relationship and genetic diversity of tree peony (Paeonia sect. Moutan) in Hunan province, the DNA fingerprints of 47 tree peony germplasm materials of Hunan province were studied using ISSR markers.
Seven primers selected from 100 ISSR primers could obtain high polymorphism and reproducibility bands.
The results showed that a total of 77 DNA bands were amplified, including 67 polymorphic bands, accounting for 87.01%. Genetic similarity analysis showed that Hunan tree peony germplasm resources have high genetic diversity.
Through computer software analysis, the average effective number of alleles was 1.3954, the average Nei’s genetic diversity index was 0.2480, and the average Shannon’s information index was 0.3859. The similarity coefficient of each type was between 0.5325 and 0.9610, with an average of 0.7515. This indicated a close genetic relationship between the materials.
UPGMA analysis divided 47 germplasm resources into 2 groups and 3 subgroups and distinguished cultivars from different sources.
This study revealed the genetic relationship and high genetic diversity of Hunan local tree peony from the molecular level, providing a theoretical basis to the hot and humid breeding work of tree peony.
