Investigation of germplasm in chrysanthemum cultivars with light-independent coloration

Abstract
Flower color is the most important ornamental quality for ornamental plants.
Anthocyanin is the main pigment determining the color of chrysanthemums, and its biosynthesis is sunlight-dependent.
In this study, we performed shading treatment at the bud stage of 372 potted chrysanthemum cultivars, and we observed the phenotypic change of flower color by the naked eye.
We had six cultivars that could accumulate anthocyanin under shading treatment.
According to pigment analysis, we found that in B40, the anthocyanin content increased almost two times more under the shading treatment than under control conditions.
RT-PCR analysis showed that in B40, the transcriptional levels of anthocyanin biosynthesis pathway genes, including MYB6, DFR and ANS, were not changed under the shading treatment compared with the control conditions.
However, these genes were not expressed in the light-dependent cultivar B14. We concluded there is a light independent signal transduction pathway in B40 that can induce the expression of ABP genes.
B40 is an important cultivar for breeding light-independent chrysanthemum cultivars.
Our work will also lay the foundation for the analysis of the molecular mechanism of anthocyanin biosynthesis in higher plants.
