Abstract
Sustainable agriculture in a climate change scenario is crucial for apple crop production.
Thus, enhancing genetic diversity and sustainable strategies to fight plant diseases are essential for maintaining agricultural productivity and food security.
Marker-assisted selection (MAS) has been shown to be an effective approach for breeding disease-resistant apple cultivars.
One of the most widely spread fungal infections affecting apple crop is powdery mildew disease, caused by the fungus Podosphaera leucotricha. A promising approach to reach durable powdery mildew disease resistance in new commercial apple varieties is pyramiding resistance genes from wild relatives.
The powdery mildew resistance gene Pl1, from Malus × robusta, has been introduced by breeding in European materials as a source of resistance to this fungal disease.
So far, the AT20 SCAR marker has traditionally been used for early selection of Pl1 resistance.
However, molecular markers more tightly associated to the gene and suitable for high throughput genotyping are needed.
To develop new molecular marker with higher effectiveness in MAS for Pl1 gene, the genomic variability of the locus containing the Pl1 gene in a set of 106 whole-genome sequencing libraries from commercial and related wild apples was extracted using the web-based tool GenoVarView.
This genomic variability was filtered, and a subset of SNPs were mapped in a family segregating for Pl1, revealing candidate SNPs closer to Pl1 than the SCAR marker.
These SNPs have been proposed for marker-assisted selection (MAS) for Pl1 gene.
