FIELD PERFORMANCE TEST OF MICROPROPAGATED REHMANNIA GLUTINOSA AND VIRUS DETECTION

Abstract
Field performance test of plants derived from apical meristem culture and conventionally propagated plants of Rehmannia glutinosa was conducted at two places to demonstrate the difference of growth and yield.
The degree of virus infection was also investigated.
There was no significant differences in length and width of leaf and number of leaves per plant between tissue cultured plants and conventionally propagated ones but chlorophyll content was increased in tissue cultured ones.
Percent of sprouting from planted rpot segments was significantly increased in tissue cultured plants and resulted in yield increase of more than 200% per 10 a.
Root thickness of tissue cultured plants at time of planting influenced percent of sprouting and yield.
Optimum root diameter ranging from 3 to 6 mm gave good yield.
Monitoring virus infection with N, tabacum and C. amaranticolor as indicator plants showed 100% with vegetatively propagated and introduced plants from China, whereas plant obtained from apical meristem showed 0% and 40% to 45% with in vitro plantlets and 1 year old plants in vivo, respectively.
Tobamovirus and unidentified virus particles were detected using electron microscope.
