Abstract
In vitro germination and tube growth of Polygonatum odoratum pollen were drastically promoted when the floral organ was added to the standard agar-sugar medium (basal medium) before sowing pollen grains.
Addition of boric acid or calcium nitrate to the basal medium promoted pollen germination a little.
The attempt of purification of the floral organ extracts showed that the substance(s) promoting pollen germination and pollen tube growth was contained in amphoteric and polar neutral fraction.
The optimal pH range of medium for germination was 4.7 to 6.1, but addition of several amino acids to the basal medium promoted pollen germination, but those amino acids did not promote pollen tube growth as addition of a floral organ did.
Thus, the promoter(s) in the floral organs can be some saccharide(s) except sucrose, some amphoteric substance(s) except the amino acids tested, or both.
