GENETIC DIVERSITY OF THREE CULTIVARS AND THEIR DESCENDANTS FROM DEGENERATIVE SEEDS IN LOQUAT ANALYZED BY RAPD MARKERS

Abstract
Genetic diversity of ‘Dawuxing’, ‘Longquan No.1’, ‘Longquan No.5’ loquat (Eriobotrya japonica (Thunberg) Lindl.) which were seed parents, and 57 lines from degenerative seeds from these three cultivars resulting from controlled pollination were analyzed by RAPD markers.
A total of 334 clear bands were generated by 23 primers, in which 319 ones were polymorphic (95.7%); Results were analyzed by POPGENE 1.31. The average values of effective number of alleles, Nei’s gene diversity, and Shannon’s information index were 1.5537, 0.3200 and 0.4812, respectively.
The similarity coefficients among the 60 loquat accessions ranged from 0.5100 to 0.9169. These loquat accessions were divided into three groups by unweighted pair group method (UPGMA) based on Nei’s genetic distances.
The genetic relationship among the 60 loquat accessions agreed with their origin.
Results indicated that degenerated seeds could be an alternative to provide diverse germplasm resources of loquat for breeding.
