BIOTECHNOLOGICAL APPROACHES FOR IDENTIFICATION OF THE MAIN VIRUSES IN LILIUM

Abstract
Virus diseases represent some of the most important diseases of Lilium, because of the devastating effects caused to the crops and the absence of effective treatments.
The viruses causing the most significant damage in Lilium are: LSV (Lily symptomless virus), CMV (Cucumber mosaic virus) and TBV (Tulip breaking virus). Other viruses identified in Lilium are ArMV (Arabis mosaic virus), INSV (Impatiens necrotic spot virus), and TSWV (Tomato spotted wilt virus) A single virus may not cause severe damage on the cultivation when alone, but if additional viruses are present, the symptoms can appear serious.
Production of virus-free propagation material is very important in nursery production.
The application of a fast and effective diagnostic technique for early detection is very important.
The aims of this research were to develop fast and reliable protocols for molecular detection to prevent the spread of the principal viruses.
At CRA-VIV, in collaboration with the ARSIA laboratory, several protocols to identify some viruses present on commercial cultivars of Lilium bulbs were developed.
The techniques for molecular diagnosis were performed on the following virus: LSV, CMV, SLRSV (Strawberry latent ringspot virus), CLRV (Cherry leaf roll virus), TSWV (Tomato spotted wilt virus), ArMV and INSV. A TaqMan® RT-PCR assay was carried out for detection of housekeeping gene18S rRNA. Tests were performed using bulbs and leaves of three LA hybrids lily cultivars, ‘Brindisi’, ‘Courier’ and ‘Lithouwen’.
