EFFECTS OF AUXIN AND CYTOKININ ON REGENERATION FROM LEAVES AND SCALES OF LILIUM × FORMOLONGI ‘RAIZEN NO.1’ IN VITRO

Abstract
The effects of plant growth regulators on regeneration from leaves and bulb scales in Lilium × formolongi Raizen No.1 were studied by in vitro culture and the content of endogenous hormones in various parts of leaves and scales was determined by ELISA. The results showed that addition of auxin and cytokinin was crucial to regeneration.
The effect of NAA at 0.5~1.0 mg/L was better than that of 2,4-D. The effect of 6-BA was better than KT, ZT or TDZ and the optimum concentration was 2.0 mg/L. The best media for regeneration from leaves and bulb scales in vitro were MS+6-BA 4.0 mg/L+NAA 0.5 mg/L, MS+6-BA 1.0 mg/L+NAA 0.5 mg/L respectively.
The induction rate (91.11%), the multiplication coefficient (7.18) and the differentiation potentiality (6.54) of bud regenerated from bulb scales were better than those from leaves (80.00%, 3.79 and 3.03). The bulb scales and basal leaves were valuable to induce buds.
This may be the result of different distribution of endogenous hormones.
There were more IAA and less CTK in the leaves than those in bulb scales, the ratio of CTK/IAA in bulb scales was higher than that of leaves.
The IAA in leaves was higher near the bract, then decreased gradually to the base of the stem, while the CTK was higher at the middle than others.
Both IAA and CTK were higher in the outer scales than the middle and inner scales.
The white and thick roots were formed on MS+IBA 1.0 mg/L, their rooting rate was up to 100% with 6.13 roots per shoot.
