SCREENING OF GENES TO IDENTIFY ARISAEMA (ARACEAE) ACCESSIONS BASED ON SINGLE NUCLEOTIDE POLYMORPHISMS

Abstract
The jack-in-the pulpit, Arisaema Mart. (Araceae), has a distinctive floral morphology characterized by a fleshy spadix surrounded by a leaf-like spathe.
Populations of Arisaema species were observed at Jeju, Korea, and some of the individuals have intermediate leaf morphologies between A. ringens, A. heterophyllum and A. amurense. The objective of this study was to evaluate 7 chloroplast and nuclear genes in four selected Arisaema accessions.
After genetic screening, we then selected A. triphyllum 26S ribosomal RNA and A. tortuosum phytochrome C-like (phyC) nuclear ribosomal genes and A. nikoense var. austral chloroplast DNA intergenic spacer region and tested all 11 accessions of Arisaema. Using sequence analysis data, clustering, based on p-genetic distance, was analyzed using the unweighted pair group method with averages (UPGMA), and the robustness of the dendrograms was tested by applying bootstrap (BS). There were two major clusters; the first major cluster divided into two sub-groups including A. kishidae, A. thundergii, A. heterophyllum, two accessions collected from Jeju (6 and 19) and another sub-group containing A. ehimense and A. yamatense. The second major cluster consisted of A. ringens, two accessions collected from Jeju (16 and 22) and one unidentified species with variegated foliage.
