From branches to FISH: a novel protocol of cytogenetic slide preparation for citrus applicable for in situ hybridization

Abstract

Cytogenetics plays an important role in applied breeding and understanding the phylogenetic relationship of citrus plants by using modern methods such as fluorescence microscopy and cytogenetic markers.
The purpose of the present research is to develop a highly efficient method for citrus chromosome slides preparation.
A roots collecting method was developed and introduced to our work on different types of citrus plants from our collection, which allows regular harvesting of root meristems without plant damaging.
We also modified the method of chromosome slides preparation, which is characterized by its speed and quality of the chromosome plates.
The results emphasize the importance of high-quality chromosome slides preparation for successful analysis of chromosomes and their structure, which is the basis for fluorescence in situ hybridization (FISH).
