Abstract
Embryogenic calli derived from ovaries and stamens of two Vitis vinifera L. cultivars, ‘Trincadeira’ and ‘Aragonez’ were bombarded with different shot pressures using gold particles coated with WRG 1515 plasmid encoding for the
-glucuronidase (GUS) and hygromycin (HPTII) genes.
Two days after bombardment, the GUS activity, evaluated by histochemical analysis, revealed 1350 and 1550 psi as the best pressures.
Bombarded calli showed a strong sensitivity to the hygromycin concentrations tested.
Embryos formation only occurred on medium devoid of hygromycin, two months after bombardment.
These embryos were then transferred to the selection medium (0.4 mg/l of hygromycin).
