DNA FINGERPRINTING OF PERSIMMON GERMPLASM USING SIMPLE SEQUENCE REPEATS

Abstract
More than 650 accessions of persimmon (Diospyros spp.) germplasm are maintained in the China National Field Genebank for Persimmon (NFGP). Accurate and unambiguous identification of these accessions is essential for germplasm preservation and utilization.
Recently a set of SSRs was derived from a library enriched for AG/TC and AC/TG motif, using Chinese local persimmon ‘Mopanshi’. After the preliminary assessment on allelic variation of each SSR in 12 genotypes, ten pairs of primer were chosen to test on 51 accessions of persimmon.
SSR fragments were produced with all primer-accession combinations, and no more than 6 fragments were amplified for each accession/primer pair combination, because most of persimmons are hexaploid.
Heterozygosity (calculated as the number of heterozygous genotypes divided by the total number of genotypes) values ranged from 0.088 to 0.971 with a mean value of 0.680, while polymorphic information content values ranged from 0.482 to 0.888. The power of discrimination values is high, ranged from 0.301 to 0.933. Based on the present data, all accessions in the NFGP collection should be fingerprinted using more SSR markers to identify individuals.
